Same problem on a dell vostro 3750

I have all the features (autostart, disable touch when... , enable touchpad at start) ON.
When I start session with no mouse connected, touchpad is disabled and shortcut doesn't work.

My temp. solution:
1) unchecked "disable touchpad when mouse plugged"
2) in gconf-editor:
apps > metacity > global_keybindings > run_command_1: <Control><Alt>0
apps > metacity > keybinding_commands > command_1: python /usr/share/touchpad-indicator/change_touchpad_state.py

With these settings I can turn on/off touchpad with the shortcut "<Control><Alt>0".
Unfortunately, when "disable touchpad when mouse plugged" is checked the python command does not work (that's why I unchecked that option).
Could spend some time to read the code, but to me this partial solution is enough, by now :)
