My system is an upgraded Ubuntu 10.04 (from 09.04).
I had the same error behaviour: cups did not start with boot time (allthough inserted in /etc/rc.local) but did start manually when entering /etc/init.d/cups start in the terminal
I found the follwoing solution:
when downgrading the package "ifupdown" from Version 0.7~alpha (which was installed during the upgrade) to Version 0.6.8ubuntu29, cups now starts automatically again.
