Something on your machine is interfering with calibre, most likely an
antivirus/security type program. Also make sure that the calibre library
folder is not on a networked drive or being simultaneously synced by a
file sync program like DropBox. Start by rebooting in safe mode, the
error should no longer occur confirming it is something interfering with
calibre.
