I'm experiencing this as well, after an upgrade. rhythmbox doesn't play the first time I hit the play button or double-click a file. The play button is toggled, but playback doesn't start. If I click it twice or double-click the file again it works fine.
Everytime playback is stopped completely (end of playlist for instance) I get this bug and need this workaround.
