Forgot to specify at the beginning, in case it was not clear: the device is the ASUS Transformer TF101 with Android Honeycomb 3.0.1

Your device has to be in disk mode for calibre to de able to detect
and connect to it. That means it has to be available as a disk to
your operating ssytem. On windows, the device must show up as a
drive with a drive letter like K:\\. Many devices have a setting
somewhere in them that causes them to connect in disk mode. Find
that setting and set it, before trying to connect your device to
calibre.

I made some more investigation: it seems that Android Honeycomb devices only mount using something called Media Transfer Protocol... Windows does not see them as proper mass storage devices at all and it does require specific drivers for each device.

Under linux it seems it should be possible to emulate an usb device using a program called mtpfs, and manipulating device rules, as explained in this post regarding the Motorola XOOM:

I've yet to try this, however, reading the thread it seems to be a bit of an unreliable solution.

If as I guess bypassing this obstacle is impossible for now, maybe you could add some notes about this to spare other Honeycomb device users a lot of searching...

calibre cannot cnnect to MTP devices. You can use the calibre Content Server
to getbooks from calibre onto your device, see
http://manual.calibre-ebook.com/faq.html#can-i-access-my-app-books-using-the-web-browser-in-my-kindle-or-other-reading-device
