First try some of the basic tips in https://help.ubuntu.com/community/SoundTroubleshooting. If you are still without sound, follow the procedure in https://help.ubuntu.com/community/SoundTroubleshootingProcedure in order to get the latest release of ALSA and for help in posting a complete bug report.
Answer the next questions:
Was it suddenly?
Do you use drivers from system > administration > hardware drivers?
What happen if you try the live CD of the current Ubuntu version (10.10) and the development version (11.04)?
