I have multiple version of python install (2.7.18 & 3.10.11). However when I run
$ sudo emacs /usr/bin/duplicity
I change the first line from #!/usr/bin/python --> #!/usr/bin/python2

Duplicity in 20.04 will expect to run against python3, I don't think changing to python2 will help.

If you run "duplicity --version" in your console, does it print a reasonable thing?

You might also try the deja-dup snap, which bundles its own duplicity and should work for you.
