not sure about specifics but when there are enough harbor spots in use it is sometimes impossible to send an expedition to the remaining spot depending on the amount of neighbor harbor spots in use counting from the start harbor location probably.
load the savegame as player 0 and you wont be able to send an expedition to the empty spot

I've tested it and the "bug" is caused by the restriction of new harbors next to enemy borders. But in most cases you should be able to send a ship to this spot because it lays in your country, means it should not be affected but it is.
